Functional photoacoustic microscopy of pH.

چکیده
pH is a tightly regulated indicator of metabolic activity. In mammalian systems, an imbalance of pH regulation may result from or result in serious illness. In this paper, we report photoacoustic microscopy (PAM) of a commercially available pH-sensitive fluorescent dye (SNARF-5F carboxylic acid) in tissue phantoms. We demonstrated that PAM is capable of pH imaging in absolute values at tissue depths of up to 2.0 mm, greater than possible with other forms of optical microscopy.

PACM: Photoacoustic Computed Microscopy
Photoacoustic Computed Microscopy (PACM) is a new label-free microscopic method that combines current photoacoustic microscopy technique with a modelbased inverse reconstruction algorithm to provide functional images of microvasculature. This article demonstrates its in vivo imaging ability of quantifying important functional parameters at the small vessel level in a rodent model. This new tech...

Recently, we developed a reflection-mode relaxation photoacoustic microscope, based on saturation intensity, to measure picosecond relaxation times using a nanosecond laser. Here, using the different relaxation times of oxygenated and deoxygenated hemoglobin molecules, both possessing extremely low fluorescence quantum yields, the oxygen saturation was quantified in vivo with single-wavelength ...

Presented here is a novel method of in vivo pH sensing utilizing a hybrid optical imaging technique, photoacoustic imaging (PAI), and pH sensitive polymeric nanoprobes. Nanoprobes with hydrophobic core containing a pH sensitive dye were synthesized and used to measure the pH level ex vivo first and then in vivo by performing experiments on a rat joint model, with an achieved precision of less t...

Commercially available high-resolution three-dimensional optical imaging modalities-including confocal microscopy, two-photon microscopy, and optical coherence tomography-have fundamentally impacted biomedicine. Unfortunately, such tools cannot penetrate biological tissue deeper than the optical transport mean free path (approximately 1 mm in the skin). Photoacoustic tomography, which combines ...

We report the development of functional photoacoustic microscopy capable of video-rate high-resolution in vivo imaging in deep tissue. A lightweight photoacoustic probe is made of a single-element broadband ultrasound transducer, a compact photoacoustic beam combiner, and a bright-field light delivery system. Focused broadband ultrasound detection provides a 44-μm lateral resolution and a 28-μm...
